サイクリンE1(英: cyclin E1)は、ヒトではCCNE1遺伝子によってコードされるタンパク質である[5]。
機能
[編集]CCNE1遺伝子によってコードされるサイクリンE1タンパク質は、高度に保存されたサイクリンファミリーに属する。このファミリーのメンバーは、細胞周期を通じてその存在量に劇的かつ周期的な変化が生じることで特徴づけられる。サイクリンはCDKの調節因子として機能する。さまざまなサイクリンはそれぞれ異なる発現と分解のパターンを示し、有糸分裂の各イベントの時間的な調整に寄与する。サイクリンE1はCDK2の調節サブユニットとして機能し、CDK2の活性は細胞周期のG1期からS期への移行に必要である。サイクリンE1はG1期とS期の境界時点で蓄積し、細胞がS期を進行するにつれて分解される。CCNE1遺伝子の過剰発現は多くの腫瘍でみられ、ゲノム不安定性をもたらすことで腫瘍形成に寄与している可能性がある。サイクリンE1はNPATと結合し、そのリン酸化に関与していることが知られている。NPATは細胞周期によって調節されたヒストン遺伝子発現に関与しており、Rbタンパク質が不在時の細胞周期の進行の促進に重要な役割を果たすタンパク質である[6]。
